Synthesis and Immunological Characterization of Toll-Like Receptor 7 Agonistic Conjugates

چکیده
Activation of toll-like receptors (TLRs) on cells of the innate immune system initiates, amplifies, and directs the antigen-specific acquired immune response. Ligands that stimulate TLRs, therefore, represent potential immune adjuvants. In this study, a potent TLR7 agonist was conjugated to phospholipids, poly(ethylene glycol) (PEG), or phospholipid-PEG via a versatile benzoic acid functional group. Compared to the unmodified TLR7 agonist, each conjugate displayed a distinctive immunological profile in vitro and in vivo. In mouse macrophages and human peripheral blood mononuclear cells, the phospholipid TLR7 agonist conjugate was at least 100-fold more potent than the free TLR7 ligands, while the potency of PEG-phospholipid conjugate was similar to that of the unmodified TLR7 agonist. When administered systemically in mice, the phospholipid and phospholipid-PEG TLR7 conjugates induced prolonged increases in the levels of proinflammatory cytokines in serum, compared to the unmodified TLR7 activator. When the conjugates were used as adjuvants during vaccination, only the phospholipid TLR7 agonist conjugates induced both Th1 and Th2 antigen-specific immune responses. These data show that the immunostimulatory activity of a TLR7 ligand can be amplified and focused by conjugation, thus broadening the potential therapeutic application of these agents.

P-33: Expression of Toll-Like Receptor 2-3 Genes in Human Sertoli Cells
Background: Toll-like receptors (TLRs) constitute a major part of innate immunity, which can distinguish pathogen associate molecular pattern. Sertoli cells create a special immunological niche that protects somniferous tubules from auto antigens and pathogens. These cells are the only somatic cells in somniferous that protect testis cells against pathogens. The purpose of this study was to eva...
متن کاملP-82: The Role of Toll-Like Receptor 2 in Ectopic Pregnancy
Background: The innate immune system is our first line of defense against invading pathogens. Toll–like receptors (TLRs) have been identified as a key role in mediating the function of innate immune system which bridges the gap between innate and adaptive immunity. For example, TLR2 is able to act as a homodimer to recognize lipoteichoic acids of Gram-positive bacteria but as a heterodimer with...
متن کاملP-189: Gene Variations of Toll-Like Receptor 2 in Endometriosis
Background: Endometriosis, a common, benign, estrogendependent and chronic inflammatory condition, is characterized by the ectopic growth of endometrial tissue that is found primarily in the peritoneum, ovaries and rectovaginal septum. In addition to its usual description as a hormonal disorder, some genetic, immunological and environmental factors can affect this disease. Toll-like receptors (...
متن کاملMonocyte Expression of Toll-like Receptor-4 in Patients with Stable Angina Undergoing Percutanoeus Coronary Intervention
Background: Toll like receptors (TLRs) are well recognized players in inflammatory conditions. Among them TLR-4 is involved in chronic inflammatory processes such as formation of atherosclerotic plaques. Objective: The present study was aimed to examine the effects of percutanoeus coronary intervention (PCI) as a revascularization method on monocyte expression of hTLR-4 and on the serum levels ...
متن کاملToll Like Receptor-4 896A/G Gene Variation, a Risk Factor for Migraine Headaches
Background: The pathogenesis of migraine involves immune-mediated mechanisms in the vascular endothelium. Toll like receptor 4 (TLR-4) is a signaling receptor of innate immunity which plays a role in various neuropathologies related to neuron inflammation. Objective: This case/control study is aimed to investigate whether TLR- 4 896A/G variation is related to migraine headaches in an Iranian po...
